Morton will be without new signings Sean Crighton and Stefan McCluskey against Spartans.
Crighton must serve a suspension carried over from his time with Elgin while McCluskey is out with a dislocated shoulder. Elsewhere, Fouad Bachirou is a major doubt with a hamstring injury.
Morton manager Jim Duffy takes charge of the Cappielow men for the first time in a competitive game, and he said: "It would be great to get my Morton career off with a win, but I watched Spartans recently and they have both quality and quantity in their squad.
``Errol Douglas, who was with Stenhousemuir last season, is a real threat and he will need watched carefully.``
Lowland League visitors Spartans, who faced Queen of the South when they took part in this competition for the first time 12 months ago, will be without the suspended Kevin Motion and John Grant .
However, new recruits Mike Bruce, Iain Thomson and Douglas all make the trip to Greenock as the Edinburgh side look to spring a surprise.
``This is a great game for us to be involved in as there is no pressure on us. We are away from home against a full-time side and no-one is expecting a Spartans win, said manager Douglas Samuel.
