New signing Soufian El Hassnaoui will be Hearts' only absentee against Annan at Tynecastle.
El Hassnaoui is nursing a groin problem but seven other new players are expected to be involved as the Edinburgh side embark on their first ever tie in this competition.
Head coach Robbie Neilson has warned his squad that the Galabank men should not be underestimated.
He said: "Annan are a more than decent side and we need to put on a good performance to get through. We have to go about things in the right manner, and by that I mean that we have to be competitive from the very first whistle."
Annan will include new signing Steven Logan if international clearance comes through following his move from Newcastle.
Manager Jim Chapman has doubts over Gregor Anderson and Kenny MacKay while goalkeeper Craig Murray has been added to the group after shot-stopper Kenny Arthur's decision to retire.
Chapman said: "This is a terrific game for our players and our club. Rather than to be worried about it I want them to embrace it.
``They will be facing a good side in front of a big crowd in a great stadium so they should enjoy it as days like that do not come along too often for part-time players."
